Can one card be in two binders at once?

No — each card entry lives in one binder, or in none. Moving it to a different binder takes it out of the old one.

It still counts towards any master set for its set regardless of which binder it's in. And if you genuinely own two copies, add it twice — then each copy can live wherever you like.
